Class Objectives /Class Outline |
This course enables students to master their skills in biochemical experiments through experiments in protein purification, fractionation of organelles, and enzymatic analysis. Basic experiments in molecular biology, such as transformation and PCR, are also carried out. Students will develop their abilities to handle various biochemical samples, such as proteins and nucleic acids. |
Class Schedule |
Lesson 1: Introduction to biochemical experiments (Ishida, Ishihara) Lesson 2: Protein purification by ion-exchange chromatography Protein assay (Ishida, Ishihara) Lesson 3: Electrophopresis of Proteins (Ishida, Ishihara) Lesson 4: Protein assay (Ishida, Ishihara, Oguro) Lesson 5: Fractionation of various organelles from rat liver (Ishida, Ishihara) Lesson 6: Protein assay of the fractionated organelles (Ishida, Ishihara) Lesson 7: Enzyme activity assay (1) (Ishida, Ishihara) Lesson 8: Enzyme activity assay (2) (Ishida, Ishihara) Lesson 9: Introduction to molecular biology and preparation of reagents and buffers (Hirano, Ishida, Ishihara) Lesson 10: Transformation of E. coli (Hirano, Ishida, Ishihara) Lesson 11: Estimation of transformation efficiency, PCR, preparation of agarose gels (Hirano, Ishida, Ishihara) Lesson 12: Agarose gel electrophoresis of PCR products (Hirano, Ishida, Ishihara) Lesson 13: Preparation of plasmid DNAs (Hirano, Ishida, Ishihara) Lesson 14:Digestion of the plasmids by restriction enzymes (Hirano, Ishida, Ishihara) Lesson 15: Analysis of the digests by agarose gel electrophoresis (Hirano, Ishida, Ishihara) |
